特丁基对苯二酚在3-氯-1,2-丙二醇影响子代雄性大鼠生殖系统发育中的作用研究
Effects of tert-butylhydroquinone on reproductive system development of male offspring rats by 3-chloro-1, 2-propanediol
目的:探讨特丁基对苯二酚(tert-butylhydroquinone,tBHQ)对亲代3-氯-1,2-丙二醇(3-chloro-1,2-propanediol,3-MCPD)喂养后的子代雄性大鼠生殖系统发育的影响。方法:孕大鼠15只按数字随机法分为对照组、3-MCPD组和tBHQ组(3-MCPD+tBHQ)。除对照组外,其他二组大鼠自孕中期(孕7 d)开始每日灌胃3-MCPD(5.0 mg/kg),同时在tBHQ组的饲料中加入质量分数1%的tBHQ喂养。孕大鼠产下子代后,取子代雄性大鼠继续喂养,测量肛门生殖器之间距离,记录睾丸下降时间。在仔鼠性成熟时,检测血睾酮浓度、睾丸及附睾质量。病理切片观察附睾中精子活力、生精上皮、曲细精管情况。蛋白质印迹法检测BAX、BCL-2蛋白。结果:3-MCPD组与对照组比较,子代雄性鼠肛门生殖器距离缩短[(4.1±0.5)mm比(5.3±0.4)mm, P<0.001];睾丸下降时间延迟[(25.5±0.7)d比(17.5±1.7)d, P<0.001];睾丸脏器指数(睾丸质量/大鼠质量)和附睾脏器指数(附睾质量/大鼠质量)降低( P<0.001)。病理结果显示3-MCPD组睾丸生精上皮及曲细精管损伤,蛋白质印迹法检测结果显示BAX升高、BCL-2降低,而对于tBHQ组各检测指标均得到改善。 结论:tBHQ对亲代3-MCPD染毒后的子代雄性大鼠生殖系统发育有保护作用,可以拮抗3-MCPD对子代雄性大鼠生殖系统发育的负面影响。
更多Objective:To explore the effect of tert-butylhydroquinone (tBHQ) on 3-chloro-1,2-propanediol (3-MCPD) in reproductive system development of male offspring rats.Methods:A total of 15 pregnant rats were randomized into three groups of control, 3-chloro-1, 2-propanediol (3-MCPD) and tBHQ intervention (3-MCPD+tBHQ). Except for control group, the other two groups received a daily dose of 3-MCPD (5.0 mg/kg) since the second trimester (7 days of gestation) and intervention group was fed with 1% tBHQ. After pregnant rats gave birth, male offsprings were taken for further feeding and anogenital distance and days of testicle descent recorded. Blood testosterone concentration was detected when young rats became sexually mature. Harvested post-decapitation, testis and epididymis were weighed. Sperm motility, spermatogenic epithelium and convoluted tubule in epididymis were observed by pathological section and BAX and BCL-2 proteins detected by Western blot.Results:Compared with control group, anogenital distance of male offspring mice shortened in 3-MCPD group [(5.3±0.4) vs (4.1±0.5) mm] ( P<0.001) and time of testicular descent was delayed [(17.5±1.69) vs (25.5±0.71) day, P<0.001]. Testicular organ index (testicular mass/rat mass) and epididymal organ index (epididymal mass/rat mass) declined ( P<0.001). Pathological examinations revealed damage of testicular spermatogenous epithelium and convoluted spermatogenous duct in 3-MCPD group. Western blot showed a rise of BAX and a decline of BCL-2. The detection indices of tBHQ group improved. Conclusion:tBHQ has protective effect on reproductive system development of male offspring rats dosed with 3-MCPD, indicating that tBHQ can antagonize the negative effect of 3-MCPD on reproductive system development.
