Abstract: Objective To establish an high performance liquid chromatography-quantitative analysis of multicomponents by single-marker(HPLC-QAMS)method for simultaneous determination of yringin,chlorogenic acid,sinapaldehyde glucoside,co-niferyl alcohol,rutoside,kaempferol-3-O-rutinoside,isochlorogenic acid B,3,5-dicaffeoylquinic acid and 4,5-diffeoylquin-ic acid in She medicine Shushen(Dendropanax dentiger),and to evaluate its quality by multivariate statistics analysis and weigh-ted technique for order preference by similarity to ideal solution method(TOPSIS).Methods HPLC-QAMS method was developed on a Waters Xbridge C18 column with acetonitrile-0.05%formic acid as mobile phase for a gradient elution,and the detection wavelength was set at 260 nm.Using kaempferol-3-O-rutinoside as a reference,the relative correction factor(RCF)of the in-ternal reference and the other eight components to be tested was established,and the RCF durability and chromatographic peak po-sitioning were investigated.At the same time,the results were compared with those measured by the external standard method to verify the accuracy and reliability of the HPLC-QAMS method.Principal component analysis(PCA),orthogonal partial least squares-discriminant analysis(OPLS-DA)and other multivariate statistical analysis and weighted TOPSIS method were used to analyze the correlation of the content results of 9 components by HPLC-QAMS method.The main potential markers affecting the quality of She medicine Shushen(Dendropanax dentiger)were excavated,and the comprehensive quality evaluation method of She medicine Shushen(Dendropanax dentiger)was established.Results The linear range of the 9 components were 3.27-81.75 μg/mL,9.85-246.25 μg/mL,0.43-0.75 μg/mL,0.31-7.75 μg/mL,1.58-39.50 μg/mL,0.59-14.75 μg/mL,1.26-31.50 μg/mL,4.55-113.75 μg/mL and 1.98-49.50 μg/mL.The average recoveries were 96.82%-100.07%(RSD<2.0%).There was no significant difference in the determination results of HPLC-QAMS method and external standard method(ESM)(P>0.05).The results of principal component analysis showed that accumulative contribution rate of former 2 principle components was 89.589%.Chlorogenic acid,syringin,3,5-dicaffeoylquinic acid and 4,5-diffeoylquinic acid were the main po-tential markers affecting the quality of She medicine Shushen(Dendropanax dentiger).The results of weighted TOPSIS method showed that the quality of She medicine Shushen(Dendropanax dentiger)in Zhejiang was the best,followed by Jiangxi,Anhui,Hu-nan and Hubei,and Yunnan and Guizhou were in the last four places.Conclusion The established HPLC-QAMS method is con-venient and accurate,and multivariate statistical analysis and weighted TOPSIS method can be used for the comprehensive evalua-tion of the quality of She medicine Shushen(Dendropanax dentiger).
