Abstract: Objective To identify the main chemical constituents of Cynoglossum officinale L.,establish an HPLC fingerprint of Cynoglossum officinale L.,quantitatively analyze 5 components,and evaluate the quality of Cynoglossum officinale L.from different sources.Methods The chemical constituents in Cynoglossum officinale L.were qualitatively analyzed by ultra performance liquid chromatography quadrupole-time-of-flight tandem mass spectrometry(UPLC-QTOF-MSE)combined with UNIFI database.The separation was performed on an Agilent ZORBAX SB C18 column(250 mm × 4.6 mm,5 pm),with a gradient elution of acetonitrile(A)-0.2%phosphoric acid solution(B)as the mobile phase.The flow rate was 1 mL·min-1,the column temperature was 30 ℃,the detection wavelength was 354 nm,and the injection volume was 10 μL.The HPLC fingerprints of 15 batches of Cynoglossum officinale L.were evaluated for similarity,and 2 chemical pattern recognition methods were used to comprehensively analyze the differences between different sources of Cynoglossum officinale L.and quantify the 5 components.Results Totally 8 compounds were identified in the positive ion mode and 16 compounds were identified in the negative ion mode.The HPLC fingerprinting common mode of Cynoglossum officinale L.was established,and 14 common peaks were identified to designate the 5 components,with similarities greater than 0.900.The quantitative analysis of the 5 components met the methodological requirements.Conclusion The HPLC fingerprints and quantitative analysis methods of the multi-indicator components are accurate and reliable,which can provide a reference for the quality control of Cynoglossum officinale L..
