加速溶剂萃取结合亲水液相色谱柱分离测定坚果中的8种生育酚异构体

目的 建立加速溶剂萃取(ASE)结合亲水液相色谱(HILIC)柱分离测定坚果中8种生育酚异构体的方法.方法 2 g坚果样品经ASE提取富集后,用BEH Amide色谱柱(150 mm×3.0 mm,1.7 μm)分离,以90%正己烷,10%叔丁基甲基醚-四氢呋喃-甲醇(20∶ 1∶0.1,V/V)作为流动相等度洗脱,荧光检测器激发波长294 nm,发射波长328 nm检测,外标法定量.结果 4种生育酚在0.5~ 80.0 μg/ml、4种生育三烯酚在0.5~ 30.0 μg/ml之间具有良好的线性(r2 >0.998),方法检出限在0.032~0.070 mg/kg之间,方法定量限范围0.096 ~ 0.210 mg/kg,三个水平加标回收率在87.5% ~ 114.2%之间,6次重复测定相对标准偏差(RSD)均≤12%.结论 本方法具有良好的灵敏度、回收率和重复性,适用于坚果中8种生育酚异构体的含量分布的测定.

抗氧化类维生素血清水平与帕金森病程的相关性

目的:检测帕金森病患者抗氧化类维生素水平,并探讨其与帕金森病程的关系.方法:对2011年6月至2015年6月的62例自发性帕金森病患者及30例健康对照组志愿者血清类胡萝卜紊、生育酚、视黄醇水平进行检测,并根据病程不同,将患者分为早期帕金森病与晚期帕金森病,与健康对照组进行比较.同时分析相关抗氧化性维生素和UPDRS运动评分的相关性.结果:血清α-胡萝卜素水平(对照组:0.22±0.09μmol/L;早期PD组:0.13± 0.09μmol/L;晚期PD组:0.09±0.04μmol/L)和β-胡萝卜素水平(对照组:2.44± 0.54μmol/L;早期PD组:1.66:±:0.58μmol/L;晚期PD组:0.80±0.41μmol/L)在健康对照组、早期PD组、晚期PD组中逐步下降,且均有显著性差异(P<0.001).并且所有PD患者血清α-胡萝卜.紊(r=-0.362;P=O.O04)和β-胡萝卜素(r=-0.45;P<0.001)水平与UPDRS运动评分呈负相关.结论:PD患者血清α-胡萝卜素和β-胡萝卜、-素水平均明显下降,并且与病情严重程度相关,低水平血清类胡萝卜.紊可能与PD的发生和发展过程相关.

Antioxidant status following postprandial challenge of two different doses of tocopherols and tocotrienols

Objective: Tocotrienols (T3s) have been hypothesized to have greater antioxidant capacity than tocopherols (Ts) due to differences in biokinetics that affect their absorption and function.The present trial compares the antioxidant effectiveness following postprandial challenge of two different doses of α-T or palm T3-rich fraction (TRF) treatments and evaluates their dose-response effects on antioxidant status.Methods: Ten healthy volunteers were given four different doses of vitamin E formulations (268 mg α-T,537 mg α-T,263 mg TRF or 526 mg TRF) in a cross-over postprandial trial.Blood was sampled at 0,2,4,5,6 and 8 hours after meal consumption and plasma antioxidant status including total glutathione,super-oxide dismutase,malondialdehyde (MDA),ferric reducing antioxidant potential and trolox-equivalent antioxidant capacity,was analyzed.Results: Supplementation with the different doses of either α-T orTRF did not significantly improve overall antioxidant status.There was no significant difference in overall antioxidant status among treatments at the different doses compared.However,a significant dose-response effect was observed for plasma MDA throughout the 8-hour postprandial period.MDA was significantly lower after the 537 mg α-T treatment,compared to the 268 mg α-T treatment;it was also lower after the 526 mg TRF treatment compared to the 263 mg TRF treatment (P < 0.05).Conclusion: T3 and α-T demonstrated similar antioxidant capacity,despite markedly lower levels of T3 in blood and lipoproteins,compared to α-T,

Comparative transcriptomics reveals hidden issues in the plant response to arthropod herbivoresOO

Plants experience different abiotic/biotic stresses, which trigger their molecular machinery to cope with them. Besides general mechanisms prompted by many stresses, specific mechanisms have been introduced to optimize the response to individual threats. However, these key mechanisms are diffi-cult to identify. Here, we introduce an in-depth species-specific transcriptomic analysis and con-duct an extensive meta-analysis of the responses to related species to gain more knowledge about plant responses. The spider mite Tetranychus ur-ticae was used as the individual species, several arthropod herbivores as the related species for meta-analysis, and Arabidopsis thaliana plants as the common host. The analysis of the tran-scriptomic data showed typical common re-sponses to herbivory, such as jasmonate signaling or glucosinolate biosynthesis. Also, a specific set of genes likely involved in the particularities of the Arabidopsis-spider mite interaction was dis-covered. The new findings have determined a prominent role in this interaction of the jasmonate-induced pathways leading to the biosynthesis of anthocyanins and tocopherols. Therefore, tandem individual/general transcriptomic profiling has been revealed as an effective method to identify novel relevant processes and specificities in the plant response to environmental stresses.
