Characteristic analysis of skin keratinocytes in patients with type 2 diabetes based on the single-cell levels

Background::Keratinocytes play an important role in wound healing; however, less is known about skin keratinocytes in patients with type 2 diabetes mellitus (T2DM). Therefore, this study aimed to search for the transcriptional characteristics of keratinocytes at the single-cell level from T2DM patients, and to provide experimental data for identifying the pathological mechanisms of keratinocytes under pathological conditions.Methods::We performed single-cell RNA sequencing on the skin tissue from two T2DM patients and one patient without diabetes-induced trauma using the BD Rhapsody TM Single-Cell Analysis System. With the help of bioinformatics R-based single-cell analysis software, we analyzed the results of single-cell sequencing to identify the single-cell subsets and transcriptional characteristics of keratinocytes at the single-cell level, including Kyoto Encyclopedia of Genes and Genomes and Gene Ontology analyzes. Results::In this study, we found specific highly expressed signature keratinocyte-related genes. We analyzed the transcriptome of keratinocytes from experimental and control groups and screened a total of 356 differential genes, which were subject to bioinformatics analysis. Enriched pathways included oxidative phosphorylation, antigen processing and presentation, prion and Huntingtons’ diseases, bacterial invasion of epithelial cells, thermogenesis, vasopressin-regulated water reabsorption, and protein processing in the endoplasmic reticulum.Conclusions::This study revealed the characteristics of keratinocytes at the single-cell level and screened a group of differentially expressed genes related to T2DM-associated keratinocytes, oxidative phosphorylation, cytokine receptor interactions, prion diseases, and other signaling pathways.

基于云和Rhapsody的脑卒中风险管理信息平台设计

目的:对脑卒中风险进行信息化管理,为预防病发、辅助临床决策以及指导康复提供一个高效信息工具.方法:以云服务的三层架构为基础,通过将Rhapsody融入到PaaS层,将医学App融入到SaaS层,构建一个新型脑卒中风险管理信息平台.该平台由数据中心、平台管理软件、移动应用开发系统以及各类医学应用App组成.结果:通过对上海市120位患者脑卒中的风险预测,得到了较高准确率的评估,验证了该平台能够很好地支持脑卒中风险的分析与管理,可作为辅助临床决策或康复指导信息化工具.结论:该平台实现了医院、社区卫生机构以及脑卒中高危人群个体的三方联动及信息共享,为脑卒中风险管理提供了一个有力工具.

The deadly global outbreak of coronavirus disease-2019(COVID-19)has forged an unrivaled threat to human civilization.Contemplating its profuse impact,initial risk management and therapies are needed,as well as rapid detection strategies alongside treatments with existing drugs or traditional treatments to provide better clinical support for critical patients.Conventional detection techniques have been considered but do not sufficiently meet the current challenges of effective COVID-19 diagnosis.Therefore,several modern techniques including point-of-care diagnosis with a biosensor,clustered regularly interspaced short palindromic repeats(CRISPR)-associated proteins that function as nuclease(Cas)technology,next-generation sequencing,serological,digital,and imaging approaches have delivered improved and noteworthy success compared to that using traditional strategies.Conventional drug treatment,plasma therapy,and vaccine development are also ongoing.However,alternative medicines including Ayurveda,herbal drugs,homeopathy,and Unani have also been enlisted as prominent treat-ment strategies for developing herd immunity and physical defenses against COVID-19.All considered,this review can help develop rapid and simplified diagnostic strategies,as well as advanced evidence-based modern therapeutic approaches that will aid in combating the global pandemic.
